The study focused on two representative sites: Sacca Fisola, within the Venice Lagoon, characterised by high maritime traffic, and Mussel Farm, a coastal area near Cavallino-Jesolo, a former mussel farm. Both sites were monitored every six months and divided into before and after cleaning activities performed by the Seabed Robotic Cleaning Platform (autumn 2022 and spring 2023 for the Sacca Fisola site and spring 2023 for the Mussel Farm site). This innovative robotic system has been designed to remove macro-litter from the seabed. Sampling and analysis of microplastics in sediment samples were performed according to the protocol described in Frias et al. (2018), Vianello et al. (2013) and JPI Oceans protocol (2018). This methodology ensures a standardised approach to collection, processing and analysis, minimising the possibility of contamination and ensuring comparable results. Sampling took place in 2023 (March and October) and 2024 (March and August). Five replicates of sediment samples were taken using a Van Veen bucket. The first 5 cm of sediment collected with a metal spoon was transferred to 1 litre glass containers for storage. In the laboratory, each sediment sample was homogenised to ensure uniformity. A 250 g aliquot was taken from each homogenised sample, treated with 100 ml of 6-10% hydrogen peroxide to remove organic matter and left to decant for 18-24 hours. After decanting, the samples were washed with water filtered through a 38 μm sieve to remove fine sediment particles. The microplastics were then separated from the sediment by density separation by adding 750 ml of filtered hypersaline solution (density: 1.2 g/cm³) to each sample. The sample was shaken vigorously for at least one minute and allowed to settle for one hour. The supernatant containing the microplastics was then filtered through a 38 μm steel sieve. To improve the efficiency of microplastic recovery, the separation and filtration process was repeated three times for each replicate sample. Procedural controls were performed in parallel with sample processing. After extraction, the microplastics were analysed under a stereomicroscope and then counted and grouped according to shape (spheres, pellets, flat and/or irregular fragments, sheets and filaments) and colour (white, black, red, blue, green, transparent or OC - other colour). The results were expressed as the number of particles per kilogram of wet and dry sediment. In the latter case, the sediment was dried in an oven at 40°C for 24 hours, after which the amount of water lost was assessed.
